In a word, yes! Assuming you are a fully registered and licensed business this is Thai law. Immigration can visit your premises to check you are doing this and there are standard fines in place if found lacking. It may seem a tad big brother but it also has the benefit of authorities knowing who and where visitors are. This can be handy in cases of accidents or any other unfortunate happening.
The online programme is very simple to use, even if the guest has misplaced his TM6 you can still enter the rest of the data.

Thanks much for the clarification. You had assumed the case of a registered and licence business. Does it apply otherwise?

If you are not a fully reg/licensed biz then "watch your six" as the military say.

This is a top priority for the immigration/police/army right now, to fine/close/jail illegal hotel businesses...
